Abstract
Original Research
introduction :
Mass spectrometry imaging (MSI) enables spatially resolved analysis of lipid distributions within biological tissues. Desorption electro flow focus ionization (DEFFI) and matrix-assisted laser desorption/ionization (MALDI) are complementary MSI techniques that provide broad lipid class coverage. This study aimed to investigate lipidomic alterations in rat brain tissue following chronic morphine exposure and to evaluate the modulatory effects of buprenorphine treatment.
Methods:
Rat brain sections from control, morphine-treated, and buprenorphine co-treated groups were analyzed using DEFFI- and MALDI-MSI in both positive and negative ion modes. Lipid-related features were compared across experimental groups, with lipid identification based on accurate mass measurements and characteristic MS/MS fragmentation patterns. Particular attention was given to lipid class–level changes and cautious annotation of detected features.
Results:
Distinct lipid class–dependent patterns were observed among the experimental groups. Chronic morphine exposure was associated with a relative reduction in glycerophospholipid-related signals and an enrichment of sphingolipid-associated features, including sphingomyelins and ceramides. In contrast, control and buprenorphine-treated groups showed higher relative levels of glycerophospholipids involved in membrane structure and lipid homeostasis. Buprenorphine treatment demonstrated a dose-dependent tendency to normalize morphine-induced lipid alterations.
Conclusion:
The combined application of DEFFI and MALDI mass spectrometry imaging revealed opioid-associated lipid remodeling in the brain at the lipid class level. These findings highlight the value of complementary MSI approaches for studying brain lipidomics and emphasize the importance of careful lipid annotation and conservative interpretation in MSI-based studies.
