Abstract
Original Research
introduction :
Direct conversion of somatic cells such as mesenchymal cells (MSCs) into oligodendrocyte
precursors (OPCs) for neurodegenerative diseases such as MS using chemicals and growth factors has been
promising. It has been shown that Olig2 is most important factors determining the fate of oligodendrocyte
lineage cells.
Methods:
The human Olig2 coding sequence was transferred into adipose tissue mesenchymal cells along
with GFP. One week after in culture in OPC medium, the cells shape was transformed. Examining gene
expression for oligodendrocyte markers, maturation and myelination tests in vitro and in vivo were
performed on the produced cells.
Results:
OPCs morphology observed within 7 days. Marker genes for OPCs were overexpressed and
suppressed for MSCs. Maturation of the cells in culture medium achieved at 2-3 weeks. Co-Culturing with
rat dorsal root neurons showed the ability to myelinate in vitro. Transplantation of the OPCs into the
demyelinated mice brain improved behavioral performance, while brain tissue examination showed that the cells get matured and myelinating.
Conclusion:
Using overexpression of Olig2 and appropriate culture medium, induced OPCs obtained
that had function in vitro and in vivo. These cells seem can be used to restore lost function of the central
nervous system.
